Uses of Primaquine

  • Primaquine is used in managing and preventing malaria, an infectious disease transmitted by mosquitoes. It belongs to a class of medicines known as 8-aminoquinolines.

  • Additionally, Primaquine is often used in combination with a blood schizonticide such as chloroquine to target various stages of the parasites and prevent relapses. This combination therapy aids in achieving a radical cure and preventing the return of Plasmodium vivax and Plasmodium ovale malaria after leaving an endemic area.

How Primaquine works

  • Primaquine is an antiparasitic medicine.

  • It works by getting rid of the malaria infection in the tissues, preventing it from spreading into the blood. This helps stop the relapses of vivax malaria. It is also effective against the gametocytes of Plasmodium falciparum, another type of malaria parasite.

  • Primaquine is also effective against what are known as hypnozoites. These are dormant liver stages of these parasites, effectively preventing relapses from these particular types of malaria.

Overdose and Missed Dose

  • Overdosing on Primaquine may cause stomach cramps, vomiting, yellowing of the skin and the whites of the eyes, heartburn, heart problems, heartbeat rhythm, bluish skin discolouration, fatigue, difficulty breathing, shortness of breath, confusion, light-headedness, dizziness, difficulty sleeping, and pale skin. If you suspect an overdose, seek medical help immediately.
  • If you forget to take a dose of Primaquine, take it as soon as you remember. However, if it is almost time for your next dose, skip the missed dose and return to your regular schedule. Do not double the dose to make up for the missed one.

Management of Side Effects

  • Most side effects of Primaquine are temporary and should resolve once you stop taking the medicine. If you experience severe side effects or worsening symptoms, consult your doctor immediately.
  • For gastrointestinal disturbances such as nausea and vomiting, consider taking the medicine with or after meals.
  • If you experience dizziness, avoid sudden movements and rise slowly from a sitting or lying position.

Use in Driving and Operating Machinery

  • Primaquine may cause side effects such as dizziness or confusion, which can impair your ability to drive or operate machinery. Avoid these activities if you experience these side effects.
